E4OD doesn't have a trans mounted VSS. There is a rear diff mounted sensor.

Generally when you have an axle sensor/connector issue you tend to have long extended harsh shifts sometimes almost forced with throttle ranges and/or speedo errors or no speedo at all.

Does the OD light flash? How many miles on the truck?

The Manual Lever Position Sensor range (MLPS) functions can be viewed with a scan tool, and wouldn't necessarily be bad just given the symptoms, or, at the same time as a rear axle sensor failure. There is a updated MLPS (neutral safety) and newer style connector to replace the old one if it hasn't been done already.

If it the trans hasn't been serviced in quite some time, a good barometer (if all else checks out) is to drop the pan and look at the collected trash. If the fluid is burnt, full of suspended metals, and the magnet has collected large 'bits' or a high volume of, then it may be more then a service required..
